New techniques make it possible for investigators to examine and sometimes quantify various aspects of nuclear morphology and function; now they can derive clinically and biologically useful information about the nucleus. This book draws together a series of techniques which have been successfully applied to the study of the nucleus of tumour cells. These are of fundamental importance to researchers in tumour histopathology and medical oncology. Detailed reviews are given of various aspects of the morphology, ploidy and karyotypic status and function of the nuclei in the cells of tumours and preneoplastic lesions. Topics covered include nuclear morphology in tumour diagnosis, the ultrastructure of the nucleus, karyotypic analyses of solid tumours, flow cytometric assessment of nuclear ploidy and other parameters, histomorphometry of the nucleus, and in situ hybridisation.

Acidic Proteins of the Nucleus focuses on the functional role of acidic nuclear proteins in differential gene expression. Historically, these proteins are referred to as acidic in nature because they are insoluble in dilute mineral acids and their amino acid composition shows a preponderance of acidic over basic amino acid residues. After an introduction to DNA-binding proteins and transcriptional control in prokaryotic and eukaryotic systems, the subsequent chapters describe various approaches for isolating, separating, and characterizing acidic nuclear proteins. The core chapters specifically cover the isolation, fractionation, and characterization of acidic nuclear phosphoproteins, and the role of these proteins in cell proliferation, cell differentiation, and cell cycle. The last two chapters address the role of acidic nuclear protein in binding steroid hormones and in gene regulation. Each chapter contains some previously unpublished work and provides recommendations for future research. This book will be a good reference background for researchers of acidic nuclear proteins.

The present text grew out of a number of lecture courses for advanced under graduate and new graduate students in nuclear physics. They were given at summer schools in Leuven, Melbourne, and at study weeks for Dutch grad uate students which aimed to emphasize fundamental and topical aspects of nuclear physics. On occasion, part of the present text was presented to stu dents from a much wider field than just nuclear physics and also within a number of general physics colloquia, where, in addition to nuclear physicists, physicists from many other fields were present. In this respect, the intention is to present, in an amply illustrated form, the key quest ions that arise in nuclear physics. At the same time we try to show why a better understanding of the atomic nucleus is not only important in itself, but also yields essential insights into the many connections to other fields of physics. We thus concen trate on the unifying themes rather than addressing in great detail particular subfields of nuclear physics. The present project does not aim to be another comprehensive textbook on nuclear physics: Many of the detailed technical arguments that enter into the picture are not developed here as they would be in a more standard textbook. Instead they are presented using analogies, quite often with simple pictures and arguments that try to convey the general line of thinking and working in nuclear physics.

The presence of sophisticated auditory processing in mammals has permitted perhaps the most significant evolutionary development in humans: that of language. An understanding of the neural basis of hearing is thus a starting point for elucidating the mechanisms that are essential to human communication. The cochlear nucleus is the first region of the brain to receive input from the inner ear and is therefore the earliest stage in the central nervous system at which auditory signals are processed for distribution to higher centers. Clarifying its role in the central auditory pathway is crucial to our knowledge of how the brain deals with complex stimuli such as speech, and is also essential for understanding the central effects of peripheral sensorineural hearing loss caused by, for example, aging, ototoxic drugs, and noise. Ambitious new developments to assist people with total sensorineural deafness, including both cochlear and cochleus nuclear implants, require a detailed knowledge of the neural signals received by the brainstem and how these are processed. Recently, many new data have been obtained on the structure and function of the cochlear nucleus utilizing combinations of anatomical, physiological, pharmacological and molecular biological procedures. Approaches such as intracellular dye-filling of physiologically identified neurons, localization of classical neurotransmitters, peptides, receptors and special proteins, or gene expression have opened the door to novel morphofunctional correlations.

The afferent connections of the cerebellar cortex of the cat have been extensively in vestigated by Alf Brodal and his collaborators using retrograde degeneration methods. These experiments (reviewed in Larsell and Jansen 1972) established that cerebellar corti cal afferents arise from widespread areas of the brain stem and spinal cord. Brain stem nuclei shown to provide input to the cerebellar cortex included the pontine nuclei, the medial and descending vestibular nuclei, vestibular cell group x, the lateral reticular nucleus, the perihypoglossal nuclei, the paramedian reticular nucleus, the inferior olive, and the external cuneate nucleus. In addition, the red nucleus and certain of the raphe nuclei were thought to send fibers to the intracerebellar nuclei, but not to the cortex. With the advent of the horseradish peroxidase (HRP) technique, new information on the distribution and organization of cerebellar cortical afferents has recently be come available. Thus Gould and Graybiel (1976) demonstrated that afferents to the cat cerebellar cortex arise from a previously undescribed lateral tegmental cell group at the level of the isthmus and from the intracerebellar nuclei, as well as from the classic precerebellar nuclei. Moreover, these studies showed that fibers from the vestibular nuclei, previously thought to be distributed only to the flocculonodular lobe and uvula, reach widespread areas of the cerebellar cortex. Experiments by other investi gators have established that the cerebellar cortex of the cat receives afferents from cer tain of the raphe nuclei (Shinnar et al. 1975; Taber Pierce et al.

The steady-state energy distribution is obtained for a group of initially monoenergetic charged particles slowing down in a fully ionized gas. The distribution function is obtained analytically by solving the Fokker-Planck equation in energy space. With reference to thermonuclear reactions, the results obtained suggest that in some situations the contribution of the slowing-down distribution to the reaction rate may be significant.

The failure of many investigators to find more than traces of deoxyribonuclease activity in the nuclei of cells has led to difficulties in attempting to ascribe a physiological role to this enzyme in living cells. Reinvestigation in this laboratory disclosed that nuclear fractions centrifugally separated from rat liver homoge nates in isotonic sucrose or in water, containing in each case 1.8 x 10 to the -3 power M CaCl(2) and washed repeatedly in the same media, retained a considerable amount of acid DNase activity. Similarly prepared nuclear fractions using calcium-free media lost nearly all of their DNase activity. This retention of activity in the presence of calcium was shown to be neither co-sedimentation of unbroken cells or of lysosomes with the nuclear fraction nor precipi tation of soluble e in the presence of calcium. The nuclear-bound enzyme was active under conditions which were shown not to remove it from the nuclei. (Author).
